On-farm mastitis test solution SOMATIC PLUS.
Application:
On-farm mastitis test SOMATIC PLUS is ready-to-use solution used for in vitro detection of subclinical mastitis in cattle.
Description of mastitis detection principle:
Surfactant sulfonate presented in the diagnostic solution interacts with the DNA of the nuclei of milk somatic cells, which number increases in mastitis milk, and forms a clot of different densities depending on the number of somatic cells. At the same time, after adding the solution, the mixture may change its color to red-orange.
Test procedure:
Place the examination paddle with four cups so that the handle of the paddle always points towards cow's head or tail so that the udder lobes and cups of the paddle are correctly aligned.
Discard the first three streams of milk into separate container.
Take an equal amount of milk (2 ml each) from all quarters of the udder into corresponding cups of the paddle. To ensure that you have the same amount of milk in all cups, tilt the paddle vertically and let excessive milk to flow out. Avoid mixing the milk from different cups of the paddle.
Using a dispenser, add test solution to each cup in amount equivalent to the volume of the milk sample (for example 2 ml). Avoid making foam.
Mix solution and milk by gently rotating paddle in horizontal plane, swirling the mixture. After 10 seconds, check test results.
Grading and Interpretation:
Observe the change in viscosity in the cups. To interpret the results, refer to the table.

Description of visible reaction |
Possible interpretation |
Somatic cell concentration |

1 |
Mixture remains liquid with no evidence of formation of a precipitate and has uniform color |
Negative |
<100 000 |
|
2 |
A thread-like precipitate forms, wich is easy to see tipping the paddle back and forth in horizontal plane. The increase in viscosity observed during mixing may disappear. |

3 |
Prominent thread-like precipitate forms, the solution transforms to a jelly-like structure |
Distinct positive |
300 000 – 500 000 |
|
4 |
The mixture has a hard, jelly-like texture with pronounced filamentous inclusions. May change the color to red-orange |
Strong positive |
>500 000 |
As a general rule, the viscosity increases in proportion to the concentration of somatic cells in the sample.
In case when inflammation has just begun and interpretation of the results is difficult, it is recommended to repeat the test before the next milking. Please please take into account that very first trickles of milk should be used for the test, which allows to observe more intensive reaction and to see differences between healthy and slightly inflamed udder lobes.
Safety information:
When working with the test solution, follow general safety and personal hygiene requirements.
Storage conditions:
Test solution is stored in a dry place, protected from light, at a temperature of plus 2 °C to plus 35 °C.
Shelf-life:
Shelf life is 24 months from the date of production.
